A single immune challenge can confirm functional activity, yet it rarely shows how broad, compartment-specific, or pathway-selective that activity is. Running separate local and systemic trials adds its own noise from differences in participants, exposure, timing, and assays.
This fifth installment in a series on early human data looks at what changes when complementary challenge models are combined within the same healthy participants. It draws on a randomised phase 1 study of two oral IRAK4 inhibitors and on further integrated designs, including cases where a null skin result did not mean absent pharmacology. It also addresses design cautions, from challenge order to multiplicity.
