The Annex 1 revision, effective since August 2023, outlines essential controls for manufacturing sterile medicinal products, emphasizing quality risk management (QRM) and contamination control strategies (CCS). Manufacturers must conduct gap assessments, remediation plans, and ensure continuous improvement to achieve compliance.
Key principles of Annex 1 include:
- Quality Risk Management (QRM): Applies to the entire document, enabling manufacturers to prioritize and mitigate risks effectively using tools like risk assessments, CAPA, and data-driven improvements.
- Contamination Control Strategy (CCS): Involves mapping end-to-end processes to identify contamination risks, implementing engineering or procedural controls, and maintaining regular reviews.
- Advancements in Technology: Encourages the adoption of isolators, robotics, and rapid detection systems.
A structured gap assessment led by subject matter experts (SMEs) is crucial for determining applicable requirements. The assessment identifies gaps, assigns corrective actions, and tracks them in the Quality Management System (QMS). Interim measures, like enhanced cleaning or gowning protocols, can demonstrate progress during remediation.
For older facilities, compliance challenges include managing pre-use post-sterilization integrity testing (PUPSIT) and upgrading infrastructure like airlocks and pass-through hatches. These must be addressed through procedural or design modifications.
QRM involves ongoing risk identification, mitigation, and monitoring. Risks must be documented, with pre-set criteria defining acceptable levels. CCS complements QRM by integrating findings into investigations, CAPAs, and QMS updates, ensuring contamination risks are minimized and monitored annually.
By leveraging expert knowledge, aligning resources, and embedding compliance into QMS processes, manufacturers can meet Annex 1 expectations and improve operational excellence
